Pronamel Science: Acid-weakened enamel surfaces can be repaired

Actively repair acid-weakened enamel with this unique formulation1,2

A unique formulation for patients with early acid erosion

Pronamel Intensive Enamel Repair contains sodium fluoride (1450 ppm) and 5% w/w potassium nitrate, as well as ingredients that can enhance the activity of fluoride to promote remineralisation and inhibit demineralisation. These include:

  • Adding lactate and controlling pH to promote fluoride uptake3,4
  • Adding a co-polymer that helps lock-in minerals, making enamel more resistant to future acid attack5
Remineralisation / demineralisation cycle

A formulation that actively repairs

The unique Pronamel Intensive Enamel Repair formulation works to promote enamel remineralisation by delivering more fluoride deep into acid-weakened enamel surfaces*2 and helps protect against acid eroision6 (inhibits demineralisation) with its low abrasivity and fluoride protection formula.

  • SEM of fluoride uptake

In vitro cross-sectional DSIMS images of enamel surface to compare penetration of fluoride with Pronamel Intensive Enamel Repair vs. two competing toothpastes.1

Adapted from GSK data on file 18005. *vs two competing toothpastes. **Regenerate Original Advanced Enamel Science (1450 ppm fluoride as sodium monofluorophosphate). Sourced in March 2018. ***Oral B Pro-Expert Gum & Enamel Repair (1100 ppm F as stannous fluoride, 350 ppm F as sodium fluoride).

  • Enamel fluoride uptake

Enamel fluoride uptake was significantly higher with Pronamel Intensive Enamel Repair vs. a competitor toothpaste and placebo (p<0.0001), as shown in a single centre, controlled, examiner-blind, randomised, three-treatment, three-period, crossover in situ design to test the remineralising performance of dentifrices.Treatment was provided once and assessed 2 and 4 hours after application. A washout phase of 2 days (using a fluoride-free dentifrice) was implemented prior to each treatment visit.2

EFU, Enamel Fluoride Uptake. Adapted from GSK Data on file 208166. *Pronamel Intensive Enamel Repair (1150 ppm F) vs. Crest ProHealth Sensitivity and Enamel Shield (1100 ppm F as stannous fluoride).

  • Surface micro-hardness recovery (SMHR)

SMHR was significantly higher with Pronamel Intensive Enamel Repair vs. a competitor toothpaste and placebo (p<0.0001).2 This was shown in a single-centre, controlled, examiner-blind, randomised, three-treated, three-period, cross-over in situ design to test the remineralising performance of dentrifices. Treatment was provided once and assessed 2 and 4 hours after application. A washout phase of 2 days (using a fluoride-free dentrifice) was implemented prior to each treatment visit.2

SMHR, surface micro-hardness recovery. SMHR measures the increase in hardness of the enamel surface due to mineral uptake i.e. remineralisation, after product use. Adapted from GSK data on file 208166. *Pronamel Intensive Enamel Repair (1150ppm F) vs. Crest ProHealth Sensitivity and Enamel Shield (1100 ppm F as stannous fluoride).

  • 3-dimensional profilometry (3DP) analysis of human enamel specimens

In a 3DP analysis of human enamel specimens to measure the loss of material (step height) between tape-protected and exposed regions of the enamel following a brushing treatment, brushing with Pronamel Intensive Enamel Repair resulted in protection from tissue loss due to demineralisation and surface wear vs. two competitors.6

Adapted from GSK data on file 181023.*vs two competing toothpastes. **Regenerate OriginalAdvanced Enamel Science (1450 ppm fluoride as sodium monofluorophosphate) Sourced in March 2018. ***Oral B Pro-Expert Gum & Enamel Repair (1100 ppm F as stannous fluoride, 350 ppm F as sodium fluoride).

  • Relative Erosion Resistance with Pronamel Intensive Enamel Repair

In a single centre, controlled, examiner-blind, randomised, three-treatment, three-period, cross-over in situ design to test the remineralising performance of dentifrices***, Pronamel Intensive Enamel Repair demonstrated significantly superior prevention of demineralisation as evidenced by % RER vs. a competitor and placebo (p<0.0001).2

RER, Relative Erosion Resistance; measures whether the dentifrices provide a combined benefit in terms of enhanced remineralisation and acid resistance of the enamel.Adapted from GSK Data on file 208166. *vs a non-optimised fluoride toothpaste. **Pronamel Intensive Enamel Repair (1150 ppm F) vs. Crest ProHealth Sensitivity and Enamel Shield (1100 ppm F as stannous fluoride). ***Treatment was provided once and assessed 2 and 4 hours after application. A washout phase of 2 days (using a fluoride-free dentifrice) was implemented prior to each treatment visit.
